Scientists Discover Why Experimental Leukemia Drug, STI-571, is Effective

A drug called STI-571, now being tested in clinics to treat a rare form of leukemia, selectively blocks a mutant enzyme that causes the disease without harming its molecular cousins. Reporting in the Sept. 15 issue of Science, a team of researchers from the Howard Hughes Medical Institute at The Rockefeller University, the Memorial Sloan-Kettering Cancer Center and the State University of New York at Stony Brook has shown how STI-571 accomplishes this feat, suggesting new avenues for the structure-based design of cancer drugs with reduced side effects. Scientists Determine 3-D Crystal Structure of Cancer-causing Protein

The three-dimensional picture of a cancer-causing protein illuminates how a mutated gene transforms cells into cancer, report scientists from the Howard Hughes Medical Institute at The Rockefeller University in the Feb. 13 Nature. The determination of this structure clarifies earlier models that sought to explain how the gene, called src, works and offers new information for designing drug therapies to fight cancers. More »
